The Mac can read and write exFat without issue. The problem is likely that the card is too new for the Built-in readers in your computers.

Luckily a new stand-alone USB reader that supports those cards is cheap.

It did not work for me formatted on OSX (I have 10.6.8) and had to use the Windows format. Someone dug up a technical fact about Mac using a blocksize Windows does not like but windows uses a blocksize OSX can handle.

But your mileage may vary.

You need to avoid having any open files on the disk. You can unmount the disk if no files are in use, in Disk Utility select the volume & use the 'Unmount' button in the toolbar. You should be able to reformat after that.

That really depends on the current partition format. You state you are changing it to a MBR partition scheme. That is different to the GUID scheme that Macs use as default. If you opt to change the partition scheme that will reformat the entire disk.

 

You can probably use a GUID partition scheme on newer Windows OS's with an ExFAT volume. That may allow you to reformat the volume instead of the entire disk.

 

Whatever you are doing it is not a good idea to reformat volumes with important data on other parts of the disk if you have no backup of the data on that disk.
